In occasione del Congresso della European Respiratory Society (ERS) 2024, il Gruppo Chiesi organizzerà due simposi dedicati all’importanza del approccio incentrato sul paziente nella gestione dell’asma e della BPCO. Il Gruppo Chiesi illustrerà inoltre il valore per i pazienti dell’uso di approcci decentralizzati innovativi nella progettazione degli studi clinici, con risultati positivi in termini di potenziale riduzione del carico sul paziente e aumento dell’accessibilità. Grazie ai recenti investimenti, il Gruppo Chiesi presenterà i principali progressi del progetto Carbon Minimal Inhalers (gli inalatori a basso impatto ambientale ), inclusi i risultati positivi di alcuni studi e l’avvio di una nuova sperimentazione clinica di Fase 3. Combinando le presentazioni degli esperti ad un interessante colloquio tra un paziente con asma e uno specialista delle vie respiratorie, il simposio si focalizzerà sul ruolo della malattia delle piccole vie aeree (SAD) nell’asma, evidenziandone l’impatto significativo sulle esacerbazioni, sulla funzione polmonare e sulla qualità di vita. Contributi dei pazienti sulla decentralizzazione di uno studio clinico sull’asma (TANGO) Il Gruppo Chiesi continua a fare progressi nel trattamento dell’asma e della BPCO con un antinfiammatorio non steroideo per via inalatoria che risponde a importanti esigenze non soddisfatte in queste patologie e che è in fase di sperimentazione nell’ambito di un ampio programma clinico internazionale, che comprende 2 studi cardine di Fase 3 sulla BPCO e uno studio di Fase 2 sull’asma. Quest’ultimo studio, denominato TANGO, rappresenta una tappa cruciale nel percorso che l’azienda sta intraprendendo per rafforzare la centralità del paziente in tutte le fasi dello sviluppo. Mentre prosegue l’arruolamento dei pazienti nella sperimentazione, Chiesi presenterà i risultati di una ricerca qualitativa condotta su pazienti con asma in 4 Paesi, in cui è stato analizzato il disegno di TANGO presentato come sperimentazione ipotetica. I risultati dello studio indicano un’accoglienza favorevole degli approcci decentralizzati, se spiegati in modo chiaro, e sono in favore di una riduzione del carico per i pazienti e una maggiore accessibilità agli studi. Sviluppi chiave del progetto Carbon Minimal Inhaler Consapevole del legame inscindibile tra cambiamento climatico e salute[1], per cui il peggioramento di uno porta all’aggravamento dell’altra[2], Chiesi condividerà i principali sviluppi del progetto Carbon Minimal Inhaler al Congresso ERS. L’azienda sta procedendo con uno studio clinico di Fase 3 a lungo termine per la nuova piattaforma di inalatori a basso impatto ambientale[3], che andrà a sostituire l’attuale propellente a base di idrofluorocarburi (HFC 134a) con un nuovo propellente a basso potenziale di riscaldamento globale (HFC 152a). Chiesi ha già completato due studi clinici a breve termine in cui è stata valutata la sicurezza del nuovo propellente[4] e studi di farmacocinetica sulle nuove associazioni fisse doppie e triple a basso impatto ambientale[5],[6]. Nel complesso, questi studi hanno fornito prove rassicuranti che confermano le prestazioni, la sicurezza e la tollerabilità simili delle nuove formulazioni rispetto a quelle attuali, consentendo una transizione agevole e sicura per i pazienti e i sistemi sanitari e riducendo al contempo l’impatto ambientale in modo significativo. Il Report di Sostenibilità 2023 del Gruppo Chiesi evidenzia gli impatti su aree strategiche come Patients, Planet, People e Prosperity, anticipando al contempo i cambiamenti normativi. Patients: Chiesi ha investito il 23,8% del fatturato in ricerca e sviluppo e si è impegnata a favore dei pazienti affetti da patologie respiratorie, sottolineando l’impatto dell’inquinamento atmosferico sulla salute. PLANET – Piano di Transizione Climatica: Raggiungere il “Net-Zero” entro il 2035 Il Gruppo Chiesi si impegna a combattere il cambiamento climatico, con l’obiettivo di raggiungere Net-Zero (Zero Emissioni Nette di gas a effetto serra) entro il 2035. Il Gruppo ha adottato un solido Climate Transition Plan (Piano di Transizione Climatica), in anticipo rispetto ai requisiti normativi previsti dalla Corporate Sustainability Reporting Directive (CSRD). Questo piano verrà utilizzato come roadmap strategica per raggiungere gli obiettivi di riduzione delle emissioni e garantirà la resilienza e l’adattabilità nel tempo nell’affrontare le sfide legate al clima. Nel 2023, rispetto all’anno di riferimento 2019, il Gruppo Chiesi ha raggiunto una riduzione del 39% delle emissioni di gas a effetto serra negli Scope 1[1] e 2[2]. Nonostante le emissioni Scope 3 siano aumentate parallelamente alla crescita dell’azienda, Chiesi sta investendo oltre €350 milioni nello sviluppo di un nuovo inalatore dosato pressurizzato (pMDI). Questa iniziativa mira a ridurre del 90% l’impronta carbonica del prodotto attuale, sostituendo il propellente oggi utilizzato con un’alternativa a basso potenziale di riscaldamento globale (GWP). Dopo il completamento di due studi clinici a breve termine per valutare la sicurezza del nuovo propellente, il Gruppo ha avviato uno studio clinico di sicurezza a lungo termine di Fase III a novembre 2023. Chiesi mira a completare lo sviluppo clinico primario del proprio portfolio di pMDI Carbon Minimal, utilizzando il propellente HFA 152a, a basso GWP, entro il 2025. La catena di valore è fondamentale nel percorso di Chiesi verso il raggiungimento del Net-Zero entro il 2035. Il Codice di Interdipendenza, sviluppato insieme a partner strategici, guida i fornitori, i partner e i distributori nello svolgimento delle proprie attività. Chiesi ha iniziato ad implementare EcoVadis come strumento principale per valutare le performance a livello ambientale e sociale dei propri fornitori, ottenendo così la medaglia[3] di platino EcoVadis nel 2023. PEOPLE – Dare Priorità al Benessere, all’Uguaglianza e allo Sviluppo della Comunità Chiesi dà priorità al benessere dei propri dipendenti e promuove un ambiente di lavoro positivo e rispettoso. Questo impegno è testimoniato da certificazioni come "Great Place to Work" (GPTW) e "Top Employer". Attraverso iniziative che promuovono le pari opportunità, un'equa retribuzione e politiche di sostegno, il Gruppo mira a creare un ambiente in cui ogni dipendente sia valorizzato nel giusto modo. Oak Hill Bio e il Gruppo Chiesi annunciano l’arruolamento del primo paziente nello studio clinico riavviato di Fase 2b finalizzato a valutare l’efficacia di OHB-607 nel prevenire la displasia broncopolmonare, la causa più comune di pneumopatia cronica nei prematuri
Il disegno di questo studio multicentrico si basa sui dati clinici del precedente studio di Fase 2a, che hanno dimostrato una riduzione significativa di casi di displasia broncopolmonare (BPD) grave OHB-607 ha il potenziale per essere la prima terapia innovativa approvata per i neonati estremamente pretermine degli ultimi 30 anni. Informazioni sullo studio di Fase 2b Lo studio clinico di Fase 2b è uno studio multicentrico, randomizzato, in aperto, a due bracci, progettato per valutare l’efficacia e la sicurezza di OHB-607 rispetto alle cure neonatali standard per prevenire la BPD e altre complicanze della prematurità nei neonati estremamente pretermine (tra le 23 e le 28 settimane di età gestazionale). Lo studio verrà condotto in diversi centri degli Stati Uniti e sarà prossimamente esteso anche al Giappone e ad alcuni Paesi in Europa. È stato progettato per coinvolgere almeno 338 neonati. OHB-607 verrà somministrato attraverso un’infusione endovenosa continua che ha inizio dopo 24 ore dalla nascita e dura fino alla trentesima settimana di età post mestruale. Tutti i bambini riceveranno contemporaneamente le cure neonatali standard in base alle rispettive condizioni e alle linee guida locali. L’endpoint primario dello studio è la riduzione dei casi di BPD grave o di mortalità entro 36 settimane di età post mestruale nei pazienti che ricevono infusione continua di OHB-607 rispetto ai neonati estremamente pretermine che ricevono solo cure neonatali standard. Ciò fornisce il razionale per valutare l’utilizzo di OHB-607 per riportare l'IGF-1 ai livelli che sarebbero stati presenti nell'utero in una gravidanza a termine, e, potenzialmente, per sostenere la crescita e lo sviluppo regolare dei polmoni e di altri organi. Chiesi e Gossamer Bio annunciano una collaborazione innovativa a livello mondiale mirata allo sviluppo e alla commercializzazione di seralutinib per il trattamento dell'ipertensione arteriosa polmonare e altre condizioni respiratorie
Una collaborazione strategica basata sull’impegno storico di Chiesi come leader nell’area respiratoria e sull’expertise di Gossamer nei processi di sviluppo nel campo dell’ipertensione polmonare. Gossamer riceverà un pagamento di 160 milioni di dollari come rimborso per lo sviluppo, con la possibilità di ottenere fino a 146 milioni di dollari in base al raggiungimento di obiettivi regolatori, e ulteriori 180 milioni di dollari legati al conseguimento di obiettivi commerciali. Si prevede una divisione paritaria dei profitti commerciali negli Stati Uniti e un accordo per la condivisione dei costi di sviluppo globale. RARE La Business Unit RARE Diseases, l'ultima nata nel portafoglio del Gruppo Chiesi, è dedicata ai trattamenti avanzati per persone affette da malattie genetiche rare o ultra-rare. Ha raggiunto un fatturato di oltre 500 milioni di euro, grazie alla forte espansione organica e all'acquisizione strategica di Amryt Pharma Plc nell'aprile 2023. Questa operazione è una pietra miliare nella storia della crescita inorganica del Gruppo, in un anno record sotto il profilo degli investimenti: ha infatti permesso a Chiesi di ampliare ulteriormente lo spettro delle malattie rare servite e di estendere il proprio impatto su un maggior numero di pazienti che necessitano di trattamenti terapeutici. Nel febbraio 2023, la FDA ha approvato la prima terapia farmacologica per il trattamento delle manifestazioni non legate al sistema nervoso centrale dell'Alfa-Mannosidosi, una malattia ultra-rara progressiva con un'ampia gamma di sintomi, in pazienti adulti e pediatrici. Nel 2023 è stata approvata in Europa, negli Stati Uniti e nel Regno Unito la terapia enzimatica sostitutiva sviluppata dal Gruppo Chiesi per il trattamento degli adulti affetti dalla malattia di Fabry. Nel dicembre 2023 è stato approvato dalla FDA anche un gel topico per il trattamento dell'epidermolisi bollosa, già disponibile in Europa e nel Regno Unito. CARE Il Franchise CARE (specialty care, neonatologia e consumer healthcare) è cresciuto in modo consistente in tutte le aree geografiche in cui il Gruppo Chiesi opera, con vendite in aumento del 2% @CER. Il surfattante naturale di Chiesi, utilizzato nel trattamento dei neonati prematuri affetti da RDS (Sindrome da Distress Respiratorio), ha continuato a eccellere, registrando una crescita del 6% @CER e riaffermando il suo ruolo di soluzione terapeutica efficace per i neonati prematuri. Alla fine del 2023, Chiesi e Oak Hill Bio hanno raggiunto un accordo per lo sviluppo, la produzione e la commercializzazione di un farmaco candidato in fase di sperimentazione clinica per il trattamento delle complicanze dell'estrema prematurità.
